Collection description

Heart failure (HF) is one of the leading causes of morbidity and mortality worldwide, with a heavy burden on global healthcare systems. Molecularly, HF is characterized by the upregulation of G protein-coupled receptor (GPCR) kinases (GRKs). These kinases regulate GPCR signaling via phosphorylation and subsequent desensitization of activated receptors. Among GRKs, GRK2 and GRK5, are the major isoforms expressed in the heart, and the levels of these kinases are elevated in human failing myocardium. Significant progress has been made in understanding the pathological role of these kinases in the heart, both as GPCR kinases and as molecules that can act independently of GPCRs, in a “non-canonical” manner. This thesis is a collection of several research studies, aiming at elucidating the role of GRKs in the modulation of cellular processes, thus providing novel insight into molecular mechanisms underlying cardiac dysfunction.
