Abstract
The thesis focuses on two main research lines. The first part investigates the Kombucha Microbial Consortium (KMC) and its ability to grow using alternative carbon and nitrogen sources, particularly agri-food by-products such as citrus residues and spent coffee grounds. Kombucha fermentation proved effective in producing bioactive molecules and bacterial cellulose. The biological activity screening showed antibacterial, anti-inflammatory, and immunomodulatory properties mainly in samples obtained from spent coffee pods using sucrose as the carbon source. Two major metabolites were isolated and identified through spectroscopic techniques (NMR and ESI-MS): 5-(hydroxymethyl) furan-2-carboxylic acid and 1-hydroxyhexane-2,5-dione. The second part of the thesis focuses on **Heyndrickxia coagulans MA-13**, a spore-forming probiotic bacterium with potential health benefits for gut health. The activity of this strain, also tested in combination with filtered kombucha beverages, was evaluated using the Twin SHIME simulated intestinal model. The results show that both kombucha-derived bioactive metabolites and H. coagulans MA-13 can modulate gut microbiota metabolic activity and functionality without significantly altering overall microbial biodiversity.
