Abstract
DOORS syndrome is a rare multisystem genetic disorder. DOORS is an acronym for the major features that characterize the disorder: deafness; onychodystrophy; osteodystrophy; mild to profound intellectual disability (previously referred to as mental retardation) and seizures. DOORS syndrome can be caused by loss-of-function mutations in the TBC1D24 gene TBC1D24 is unique in containing an N-terminal TBC domain and a C-terminal TLDc domain. However, the molecular function of TBC1D24 is still unclear. Here we investigate the mechanism underlying the neurological defects observed in TBC1D24-associated DOORS syndrome.
