Collection description

Craniofacial defects often require bone reconstruction. The patients with diabetes have increased risks of suffering from periodontitis and failures in bone augmentation techniques. The use of mesenchymal stem cells (MSCs) and of platelet-derived products have been considered in regenerative medicine procedures. Here MSCs were isolated from Bichat’s buccal fat pad (BFP) and the effect of glucose and platelet rich plasma ( PRP) were evaluated on their proliferation and osteogenic differentiation. Cells were cultured in DMEM (Dulbecco’s modified Eagle’s medium) - F12 and with 5.5 mM glucose (Low glucose; LG) or 25 mM glucose (High glucose; HG). The number of BFP-MSCs cultured in LG was significantly higher compared with those cultured in HG. In HG medium, the expression levels of ALP were significantly lower compared with LG medium. Moreover, PRP increased cell number significantly in LG respect to HG medium, but there was no more statistically significant difference in ALP mRNA levels between cells differentiating in LG and HG, suggesting that, at least in HG, PRP potentiates the differentiation capability of BFP-MSCs.
