Abstract

Cephalopods represent one of the most advanced classes of Phylum Mollusca, thanks to sophisticated sensory and cognitive abilities supported by a highly developed nervous system. Among them, Octopus vulgaris constitutes an ideal model for the study of sensory perception and its implications on animal welfare. This thesis adopts a multidisciplinary approach that integrates molecular, behavioral, and methodological analyses to contribute to the understanding of perceptual mechanisms and provide tools for the development of ethical and effective experimental protocols, in accordance with Directive 2010/63/EU. The research begins with the study of extraocular photoreception, which highlights the expression of functional opsins in suckers, optical lobes, and skin, suggesting the existence of an ability to perceive visual information in an alternative way, involved in mimicry, defensive mechanisms, and circadian regulation. Subsequently, an innovative experimental setup has been developed for the analysis of octopus responses to environmental stimuli in a controlled environment, laying the foundations for future studies on the acoustic perception of the species. At the same time, the role of suckers – as organs that are not only sensory, but also have anti-proliferative and anti-inflammatory activity – is explored in depth. In particular, the pentapeptide OctoPartenopin and its analogs show a potential role in tissue protection and repair. Finally, the work addresses aspects related to animal welfare. Analysis of capture-induced stress has revealed physiological and molecular alterations, emphasizing the need for an acclimatization period and controlled handling procedures before experimental studies. In addition, an anesthesia protocol based on the combined use of isoflurane and magnesium chloride has been validated – ensuring rapid, effective, and fully reversible anesthesia, thereby improving experimental management and animal welfare protection.
