Abstract

This PhD thesis focuses on the development and application of integrated methodological approaches to investigate microbial diversity in subsurface environments. By combining geochemical analyses of major ions and trace elements with high-resolution molecular microbiology data, this work explores how physicochemical conditions shape microbial community structure and function. A central aim of the thesis is the definition and validation of standardized analytical protocols for the measurement of major ions and trace elements in hydrothermal fluids, addressing contamination control and experimental reproducibility. The results highlight the key role of major ions and trace elements in regulating microbial metabolisms and ecosystem dynamics in extreme environments, contributing to a deeper understanding of geochemistry–microbiology interactions in the terrestrial subsurface.
