Abstract
Behavioral dysfunctions in dogs represent one of the main social concerns, since they can endanger animals and human-dog relationship. They can bring about signs of stress or fear in dogs, either in unique situations or in multiple settings, apart from the presence of scaring stimuli (human, animal, place, scent, auditory stimuli, object). Accordingly, alongside with genetic susceptibility, generalized fear can be triggered by environmental factors, traumatic experiences, and medical conditions. Behavioral therapy, context-suited management and neurochemical manipulation, through nutrition, supplements, medicines, and pheromones, are the mainstays of the currently accessible treatments. Recent and growing evidence in humans and animals highlight the importance of the gut-brain axis in the modulation of the brain physiology and behavior. Therefore, during my PhD programme, taking advantage of high-throughput approaches, we sought to investigate the impact of the generalized fear upon microbiota, metabolome and lipidomic signatures in family dogs. Microbiota analysis from the stool of the dogs diagnosed with fear showed a differential abundance of taxa belonging to Proteobacteria and Firmicutes Phyla, when compared to the healthy control group. In addition, serum metabolome profiling documented alterations of molecules associated with GABA and glutamate neurotransmission, as well as bile acids metabolism, in the case group. Finally, the patients showed altered levels of phosphatidylcholines, which are regarded as key building block of the cell membranes and relevant players of the brain function as well. Overall, our preliminary study highlighted an intriguing role for the microbiota-metabolome-lipidome network in the generalized fear, thus shedding light on a better understanding about the pathophysiology of mood-related dysfunctions affecting companion animals and paving the way for more effective therapeutical approaches.
