Abstract
This PhD thesis aimed to fully characterize a set of mother tinctures (MTs) derived from plant extracts, to define their chemical composition, and evaluate their biological activities on different experimental systems. An untargeted liquid chromatography high-resolution tandem mass spectrometry (LC-MS/MS) analysis was performed to profile the MTs. Based on these results, some extracts were further investigated for their biological activity on a panel of immortalized and cancer cell lines. Moreover, the antibacterial and antifungal activities of MTs were tested against plant beneficial and phytopathogen strains. Three additional extracts, Rhododendron tomentosum, Calendula officinalis, and Artemisia abrotanum, were further investigated for their detailed metabolomic profiles. Moreover,C. officinaliswas specifically tested for its anti-inflammatory properties. To explore innovative applications of plant extract, A. abrotanum extract was encapsulated into cloudberry nanovesicles, and a comprehensive untargeted metabolomic and lipidomic profiling was performed alongside biological assays on cell-based models. Metabolomic profiling uncovered extensive molecular diversity and led to the identification of previously undetected compounds. Biologically, the extracts exhibited a wide range of activities. Notably, Propolis, Thuja occidentalis, and Chelidonium majus are active against both beneficial and phytopathogenic bacteria. Interestingly, C. officinalis and C. majus show selective antifungal effects. Furthermore, encapsulation of A. abrotanum into cloudberry nanovesicles enhances biocompatibility and antioxidant activity. Overall, these findings expand current knowledge on the chemical and biological properties of mother tinctures and support their potential applications in several fields, including pharmaceutical and agricultural sectors, as well as in the innovative use of extracellular vesicles.
